Ubiquitin-specific protease 2 deficiency effect on the kidney

Analysis of kidneys from mutants deficient for ubiquitin-specific protease 2 (Usp2). Results provide insight into the role of Usp2 in kidney function.

Transcriptome analysis on whole kidney of Usp2 knockout mice

(Submitter supplied) Ubiquitylation plays an important role in the control of Na+ homeostasis by the kidney. It is well established that the epithelial Na+ channel ENaC is regulated by the ubiquitin-protein ligase NEDD4-2, limiting ENaC cell surface expression and activity. Ubiquitylation can be reversed by the action of deubiquitylating enzymes (DUBs). One such DUB, USP2-45, was identified previously as an aldosterone-induced protein in the kidney, and is also a circadian output gene. more...

Role of TBC1D4 in mouse kidneys: Identification of compensatory mechanisms in the DCT of TBC1D4-deficient mice

(Submitter supplied) In vitro studies identified TBC1D4 as an regulator of renal ion and water transporting proteins. However, TBC1D4-deficient mice did not show a defective renal salt and water homeostasis. With microarray analysis we aimed to decipher compensatory mechanisms in TBC1D4-deficient mice which might mask the renal phenotype already on transciptomic levels.

Effects of supernatants of USP2 knockdown HL-60 cells and mock transfected HL60 cells on differentiated 3T3-L1 cells

(Submitter supplied) Ubiquitin specifc protease (USP) 2 modulates various cellular responces including carcinogenesis, spermatogenesis, and glucose metabolism. So far, we established USP2 knockdown HL-60 cells to explore roles of USP2 in macrophages. In this experiment, we investigated whether USP2 knockdown HL-60 cells more prominently modulate function of adipocytes compared with control cells.

Effects of PMA on exon usage of HL60, THP-1, and U937 cells

(Submitter supplied) Myeloid leukemia cell lines HL60, THP-1, and U937 undergo macrophage-like differentiation after treatment with phorbol ester. To explore genes whose exon usage was altered during macrophage differentiation, we compared exome of PMA-treated (differentated) and vehicle-treated (undifferentiated) myeloid cell lines.

Effects of gene scilencing of USP2 on expression profiles of HL60 cells

(Submitter supplied) So far, we have found PMA induced USP2b isoform in myeloid leukemia cell lines such as HL60, THP-1, and U937. To explore molecular function of USP2 in the cells, we assess expression profiles of HL60-derivatives continuously expressing shRNA for USP2 and control shRNA.

Expression profile of rat kidney of fetuses with 19 days of gestation under chronic rotation of the photoperiod.

(Submitter supplied) Previously, we reported that of pregnant dams rats exposure to continues light and rotation of photoperiod have detrimental effects on the mother, fetus and adult offspring, with changes in the expression profile of fetal heart and liver. We investigated whether the chronic rotation of the photoperiod (CPS) during gestation can modify the fetal kidney expression profile at gestational day 19 by Affymetrix microarray analysis.
