Retinoid X receptor activation effect on stimulated RAW264.7 cells

Analysis of RAW 264.7 macrophages pre-treated with vitamin A derivative 9cis-retinoic acid (9cRA), then stimulated with bacterial LPS or viral mimetic polyI:C. 9cRA is a high affinity ligand for retinoid X receptor (RXR). Results provide insight into the role of RXR in host antiviral response.

Gene expression during ligands activation of RXR in activated RAW264.7 cells

(Submitter supplied) Ligands activation of RXR modulate host antivarl response. We used microarray to determine if 9cRA could regulate the antiviral gene expression in LPS- and polyI:C triggered RAW264.7 cells.

Retinoid X Receptor activation reverses the age-related deficiency in myelin debris phagocytosis and enhances remyelination

(Submitter supplied) The efficiency of central nervous system (CNS) remyelination declines with age. This is in part due to an age-associated decline in the phagocytic removal of myelin debris, which contains inhibitors of oligodendrocyte progenitor cell differentiation. In this study we show that expression of genes involved in the retinoid X receptor (RXR) pathway are decreased with aging in myelin-phagocytosing cells. more...

Genome wide characterization of a STAT1-independent antiviral and immunoregulatory transcriptional program induced by IFNβ and TNFα reveals non-canonical STAT2 and IRF9 pathways

(Submitter supplied) Interferon (IFN) β and Tumor Necrosis Factor (TNF) α are key players in immunity against pathogens as well as in the development of autoinflammatory and autoimmune diseases. Accordingly, their molecular pathways have attracted much interest as therapeutic targets. Compelling evidence has shown that the antiviral and inflammatory transcriptional response induced by IFNβ is reprogrammed by crosstalk with TNFα. more...

Next Generation Sequencing Facilitates Quantitative Analysis of VSV-stimulated Wild Type and lncRNA-ISIR -/- spleen Transcriptomes

(Submitter supplied) Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. Methods: spleen mRNA profiles of 8-weeks-old WT and lncRNA-ISIR, used to be called lnRNA-IRF3, knockout mice were generated by deep sequencing, using BGISEQ-500 system. The sequence reads that passed quality filters were analyzed at the transcriptional level with RSEM (RNA-seq by Expectation Maximization). more...

VDR ChIP-seq in human monocytic THP-1 cell line

(Submitter supplied) Treatment with calcitriol, a specific VDR agonist, augmented the heterodimerization between VDR and RXR. We also wanted to investigate its impact on VDR binding to genomic regions. ChIP-seq experiments were carried out with the human monocytic THP-1 cell line under either vehicle (1:1 DMSO-ethanol) or calcitriol (100 nM) treatment conditions. Our finding is that calcitriol may have both a stimulatory or an inhibitory effect on VDR binding depending on the presence of its specific response element (VDRE), a less specific nuclear receptor (NR) half-site or absence thereof ("None"). more...
