Title RNA-seq data showing the effects of rpoD mutation t1682c in Pseudomonas fluorescens
Organism Pseudomonas [fluorescens] SBW25
Experiment type Expression profiling by high throughput sequencing
Summary We report RNA-seq data for four Pseudomonas fluorescens SBW25 types: SBW25 (wild type) and dervied types 6A4, 6B4-Cap- and 6B4-Cap+. The aim of the study is to see the effect of a mutation in rpoD (carried by 6B4) on the transcriptome.

Overall design Method: 3x single colonies of each of SBW25, 6A4 and 6B4 were grown on KB agar, precultured in shaken KB, grown to mid-exponential phase in shaken KB flasks. Total mRNA isolated with RiboPureTM Bacteria Kit (AmbionĀ®). Note that 6B4 was separated into Cap- and Cap+ fractions by centrifugation before RNA extraction. Normalized mRNA-seq library preparation, followed by 100 bp single end sequencing, was performed by the Australian Genome Research Facility (Brisbane, Australia).
