Summary |
We screened the genome of OCI-Ly-7, a Diffuse Large B-cell Lymphoma cell line, for genes mediating the response to rituximab treatment. We utilized the genome-wide library GeCKO v2 (A+B) from Feng Zhang. (Addgene#1000000049)

Overall design |
In addition to mock treated sample, we subjected cells to two different rituximab conditions. RTX-SEC (short exposure to CDC conditions, SEC, followed by non-CDC conditions for a total of 14 days) and RTX-REC (repeated exposure to CDC conditions, REC, maintained for 21 days). The rationale was to expose cells in the RTX-SEC group shortly to CDC conditions followed by non-CDC conditions, whereas cells in the RTX-REC group were repeatedly subjected to conditions supporting CDC
