Overall design |
We cultured C. glabrata in YPD containing 0.06 µg/ml micafungin or 0.25 µg/ml caspofungin for 24 hours, harvested the cells, used fluorescence-activated cell sorting to enrich for echinocandin-tolerant cells, followed by modified single cell RNA sequencing at the Columbia University Genome Center. Untreated stationary or logarithmic phase cells were used as controls.
