Overall design |
Total RNA was purified from 5000-20000 transduced (CRLF2+ or GFP+) CD45+CD3- cells that were sorted from spleens of transplanted mice. cDNA libraries were prepared using SMARTer v3 kit (Clontech Laboratories, Inc. A Takara Bio Company, Mountain View, CA, USA) followed by the Nextera-XT indexing protocols (Illumina, CA, USA). Genome-wide expression profiles were obtained by sequencing of the samples on Illumina NextSeq 500 using NextSeq 500/550 High Output v2 kit.
