Overall design |
Three biological replicates of each genotype were used for high-throughput sequencing analysis. For seed germination-related experiments, all plants were grown side-by-side, and the seeds were harvested on the same day. The seeds were dried at room temperature for about 2 months prior to subsequent experiments. Including sterilization and plating, seeds were exposed to white light for 1 hour. For the phyB-off condition, the seeds were exposed to far-red light (5 μmol·m-2·s-1) for 5 min to inactivate phyB. For the phyB-on condition, the seeds were irradiated with red light (20 μmol·m-2·s-1) for 5 min to activate phyB. The seeds were then incubated in the dark for the indicated time, and the germination rates were determined in the dark room.
