Treatment protocol Eye imaginal disc was dissected from 3rd instar larval stage under the dissecting scope.
Growth protocol We used the GMR-Gal4/UAS system to generate transgenic flies expressing wild-type (Wt) or mutant (Mut, C96Y) human proinsulin. Flies were hemizygous for the transgene and maintained at 25 degree.
Extracted molecule total RNA
Extraction protocol Extraction of total RNA by MELTTM Total Nucleic Acid Isolation System (Ambion) and amplification of total RNA by OvationTM RNA Amplification System V2 (NuGEN) were performed according to the manufacturer's instructions.
Label Biotin
Label protocol Fragmentation and labeling process was carried out by FL-OvationTM cDNA Biotin Module V2 (NuGEN).
 
Hybridization protocol 3.75 µg of fragmented and biotin labeled targets was hybridized to array for 18 h at 45°C and 60 rpm in an Affymetrix Hybridization Oven 640.
Scan protocol GeneChip were scanned using Affymetrix GeneChip Scanner 3000 7G.

Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis setting and global scaling as normalization method.
