Cyanine-3 (Cy3) labeled cRNA was prepared from 0.5 ug RNA using the Genomic Enzymatic Labeling Kit (Agilent) according to the manufacturer's instructions, followed by RNAeasy column purification (QIAGEN, Valencia, CA). Dye incorporation and cRNA yield were checked with the NanoDrop ND-1000 Spectrophotometer.

Hybridization protocol |
0.5 ug of Cy3-labelled cDNA hybridized to Agilent SurePrint G3 Human GE Microarray (8×60K) for 17 hours in a rotating Agilent hybridization oven. After hybridization, microarrays were washed 1 minute at room temperature with GE Wash Buffer 1 (Agilent) and 1 minute with GE Wash buffer 2 (Agilent), then dried immediately by brief centrifugation.

Slides were scanned immediately after washing on the Agilent DNA Microarray Scanner using one color scan setting for 8x60k array slides.

Gene expression of CD161-nega Vdelta1 cells

Data processing |
The scanned images were analyzed with Feature Extraction Software (v.10.7.3.1, Agilent). We examined the gene variations among CD161-negative and CD161-positive samples. And, we analyzed the gene expression of CD161-positive Samples to CD161-negative Samples. We showed the normalized data using CD161-negative Samples as a baseline. In the Sample data tables (normalized data tables), $B!H(B0$B!I(B values mean $B!H(BLog2 0$B!I(B. So, the CD161-negative Samples have only $B!H(B0$B!I(B values.
