Characteristics treatment: Thioridazine
Treatment protocol For each replicate, one part was treated with thioridazine hydrochloride (final conc. 16 mg/l), one part was treated with thioridazine hydrochloride (final conc. 16 mg/l) AND dicloxacillin (final conc. 0.125 mg/l), and the remaining part was left untreated (control). During the 30 min treatment, cultures were incubated at 37 °C with agitation.
Growth protocol Three independent ON cultures (replicates) of S. aureus USA300 (grown aerobically in Mueller-Hinton broth at 37 °C) were diluted in Brain Heart Infusion broth in seperate flasks and grown to early exponential phase (OD600 0.2) at 37 °C with agitation. Each culture was then split in three parts and treated as described under treatment protocol.
Extracted molecule total RNA
Extraction protocol Samples were immediately stabilized with the RNAprotect reagent (Qiagen) and total RNA was extracted by a hot acid-phenol procedure (Moazed et al. J Mol Biol. 187:399). Genomic DNA was removed with Dnase I (Roche) and the purification procedure was repeated. RNA integrity and gDNA absence was confirmed using a 2100 Bioanalyzer (Agilent).

Data processing The raw data (.xys files) was preprocessed using the BioConductor Oligo package and R program language based on the Robust Multichip Average (RMA) method (Irizarry et al. Nucleic Acids Res 31:e15).
