Treatment protocol |
For each replicate, one part was treated with thioridazine hydrochloride (final conc. 16 mg/l), one part was treated with thioridazine hydrochloride (final conc. 16 mg/l) AND dicloxacillin (final conc. 0.125 mg/l), and the remaining part was left untreated (control). During the 30 min treatment, cultures were incubated at 37 °C with agitation.

Growth protocol |
Three independent ON cultures (replicates) of S. aureus USA300 (grown aerobically in Mueller-Hinton broth at 37 °C) were diluted in Brain Heart Infusion broth in seperate flasks and grown to early exponential phase (OD600 0.2) at 37 °C with agitation. Each culture was then split in three parts and treated as described under treatment protocol.

Extraction protocol |
Samples were immediately stabilized with the RNAprotect reagent (Qiagen) and total RNA was extracted by a hot acid-phenol procedure (Moazed et al. J Mol Biol. 187:399). Genomic DNA was removed with Dnase I (Roche) and the purification procedure was repeated. RNA integrity and gDNA absence was confirmed using a 2100 Bioanalyzer (Agilent).
