Title |
Mouse Ba/F3 cells transfected with pMSCV empty vector supplied with IL3 and treated with Glivec |
Sample type |
RNA |
|
|
Source name |
Ba/F3 cells
|
Organism |
Mus musculus |
Characteristics |
treatment: pMSCV empty vector transfection + cuture in the presence of IL3 + 4h treatment with Glivec cell type: Mouse Pro B cells cell line: Ba/F3
|
Treatment protocol |
Oncogene-transformed cells were selected for growth in the absence of IL3. Before RNA extraction, cells were left untreated or incubated for 4 hours with 100 nM Glivec. Samples were washed once with PBS before RNA extraction. In control condition, cells were cultured in the presence of IL3.
|
Growth protocol |
Ba/F3 cells were grown in DMEM medium supplied with 10% FBS, penicillin and streptomycin.
|
Extracted molecule |
total RNA |
Extraction protocol |
Total mRNA was extracted using Rneasy mini kit (Qiagen) according to manufacturer's protocol.
|
Label |
biotin
|
Label protocol |
Biotinylated cRNAs were prepared according to the standard Affymetrix protocol from 5 µg total mRNA.

Hybridization protocol |
Affymetrix standard
|
Scan protocol |
Affymetrix standard
|
Description |
Gene expression profile from Ba/F3 cells expressing control vector treated with Glivec for 4 hours and kept in culture with the presence of IL3
|
Data processing |
Affymetrix GCOS suite version 1.4.0 was used. The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
