Agent: GM-CSF and IL-4 Treatment dose: GM-CSF (800 IU/mL) and IL-4 (1000 IU/mL) Treatment time: 6 days In-vitro treatment: Elutriated monocytes were prepared from 20-80 year old healthy donors. Elutriated monocytes were plated onto 24 well plates and differentiated into moDCs with RPMI-1640 (+glutamine; Invitrogen) supplemented with 10% heat-inactivated FCS, 0.5% HEPES, 1% Penicillin/Streptomycin, and 0.01% gentamycin, in the presence of GM-CSF (800 IU/mL) and IL-4 (1000 IU/mL) for 6 days.

Affymetrix RMA Data Processing Calculation Method: After Robust Multichip Average (RMA) normalization, differentially expressed genes were identified by One-Way Repeated Measures ANOVA (p<0.01) corrected by Benjamini-Hochberg's False Discovery Rate method (p<0.05) and this gene list was further filtered for between-group alpha levels of p<0.01, p<0.005, and p<0.001 for genes differentially expressed from moDCs differentiated with GM-CSF and IL-4 or GM-CSF, IL-4 and 1 IU of IFN-gamma. The statistically significant gene expression changes were functionally analyzed using Ingenuity Pathways Analysis (Ingenuity Systems) software.
