Treatment protocol |
Pooled hypothalami from 10 mice were homogenized, centrifuged and mixed with with streptavidin coated Dynal magnetic beads previously incubated with biotinylated protein L and monoclonal mouse anti-GFP antibody. The homogenate represents the total sample. The homogenate and beads were mixed overnight followed by two washes in high potassium wash buffer then a wash in high potassium wash buffer with DTT, cyclohexamide and RNAse inhibitors. The supernatant represents the IP sample.

RNA was extracted with Agilent Absolutely RNA nanoprep kit. RNA was amplified and converted to cDNA using Ovation PicoSL WTA System (NuGen, San Carlos, CA)
