Treatment protocol |
All blood and bone marrow have been subjected to ficoll density centrifugation and frozen viable cells were prepared in DMSO+FCS+RPMI and stored over liquid nitrogen. Tissue samples have been minced and a cell suspension was prepared. Cell suspension was then stored as frozen viable cells in DMSO+FCS+RPMI and stored in liquid nitrogen

Approximately 10X106 cells per case were used for the extraction of total RNA by RNeasy Micro kit (Qiagen, Valencia, CA) according to manufacturer’s instructions.

Each RNA sample was labeled by synthesizing complementary cDNA and converted to labeled complementary RNA using T7 RNA polymerase in the presence of biotinylated UTP (Affymetrix). This cRNA is used as the target for microarray fragmentation and hybridization.
