Extraction protocol |
Total RNA was extracted by TRIzol (Invitrogen) from CD11b+ sorted cells according to the manufacturer?s instructions with little modifications. The quality and quantity of RNA sample were determined by Agilent RNA 6000 Nano Chip (Agilent Technologies). cDNA from purified total RNA was produced by M-MLV reverse transcriptase (Invitrogen) following manufacturer protocol. All the pre-hybridization quality controls were performed with the Agilent 2100 bioanalyzer (Agilent Technologies).

5 μg of total RNA were used to generate double-stranded cDNA (Invitrogen). Synthesis of Biotin-labeled cRNA was performed using the BioArrayTM HighYieldTM RNA Transcript Labeling Kit (ENZO Biochem).
