Treatment protocol SKPs proliferated as spheres. After one or two passages, spheres were placed in Trizol reagent. Schwann cells or SKP-Schwann cells were passaged once, grown until confluent, then washed and placed in Trizol reagent.
Growth protocol Dorsal trunk skin samples were cut into small pieces and enzymatically digested and filtered until single cells were isolated. These cells were cultured in proliferation-promoting conditions as previously described. The resulting SKP spheres were harvested for RNA or plated in Schwann cell growth media. Sciatic nerves were dissected and enzymatically digested to liberate single cells. These were plated and grown in Schwann cell growth media.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed with the RNeasy Mini Kit (Qiagen) according to manufacturer's instructions.
Label biotin
Label protocol Single-stranded cDNA was generated from the cRNA with the WT cDNA Synthesis Kit (Affymetrix) and then fragmented and labeled with the WT Terminal Labeling Kit (Affymetrix).
 
Hybridization protocol Samples were hybridized with GeneChip Rat Gene 1.0 ST Arrays (Affymetrix) and scanned at the HSC Microarray Facility.
Scan protocol The hybridized microarray image was scanned with the GeneChip Scanner 3000 7G (Affymetrix).
Description Gene expression data from adult rat dorsal trunk SKP-derived Schwann cells
Data processing Raw probe intensity values were background corrected, normalized with quantile normalization, transformed into the log2 scale, and summarized into probesets using the Robust Multichip Analysis (RMA) algorithm at the gene level in the Affymetrix Expression Console program.
