Treatment protocol |
Capan1 cell were transduced with amphotropic retroviruses carrying either an MSCV-SV40-PuroR or MSCV-PROX1-PuroR, for 2.5 h. Three indeendent transductions were performed for each MSCV_puro and MSCV_PROX1_puro. Two days after transduction, the cells were incubated with 0.5 ug/mL puromycin and selected for 4 days. The selections was stopped after all the cells in the growth control (non-transduced Capan1) have died.

Capan1 cells were grown in IMDM/FCS medium according to ATCC specifications.

Six (6) micrograms of cDNA was fragmented and then hybridized for 16 hr at 45C on an Affymetrix GeneChip Primeview Human Gene Expression Array. Following hybridization, arrays were washed and stained using theAffymetrix Fluidics Station 450 system.

GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G.

Data processing |
Probe signals were quantile normalized and summarized by the RMA algorithm using Partek Genomics Suite software v6.5.
