Extraction protocol |
Total RNA was extracted from fresh frozen tissues using the Roche High Pure miRNA Isolation kit (Cat No 05080576001, Roche Diagnostic, Indiana, IN, USA) in accordance with the manufacturer’s protocol. The quality and quantity of small RNAs present in the total RNA extract was assessed using an Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA).

100ng of total RNA was used. miRNAs were fluorescently labelled by lagation of one Cyanine 3-pCp molecule to the 3' end of a RNA molecule

Hybridization protocol |
Labelled material was hybridised to the arrays at 55C for 20hrs, rotating at 20rpm. Slides were washed with agilent wash buffers.

Agilent High Resolution Scanner, G2505B was used. Agilent Feature Extractiob version 10.7.3.1. software was used for data extraction and protocol used by Fe software: miRNA 107 Sep09
