Title |
Nonresponder, biological rep8 |
Sample type |
RNA |
Source name |
AB1-HA cells innoculated into mice.
Organism |
Mus musculus |
Characteristics |
tissue: AB1-HA tumor strain: Balb/c group: Nonresponder
Treatment protocol |
The mice were treated with 100 ug anti-CTLA-4 intraperitoneally on day 5 or 6. The tumor was removed 7 days later for gene expression profiling.
Growth protocol |
Balb/c mice were inoculated subcutaneously with 5 x 10^5 AB1-HA cells on both flanks on day 0.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted with TRIzol followed by Rneasy Min Elute.
Label |
biotin
Label protocol |
250ng of total RNA was amplified (Ambion WT kit), labeled with biotin (Affymetrix terminal labelling kit) and fragmented according to manufacturer’s instructions. This was performed by the Ramaciotti Centre for Gene Function Analysis (University of New South Wales).
Hybridization protocol |
5.5 μg of fragmented, biotin labeled ssDNA was hybridized to each Mouse Gene 1.0ST microarray according to manufacturer’s instructions. This was performed by the Ramaciotti Centre for Gene Function Analysis (University of New South Wales).
Scan protocol |
The samples were scanned on a GeneChip® Scanner 3000 7G (Affymetrix).
Description |
Gene expression data from nonresponding AB1-HA cells that were innoculated into mice and treated with anti-CTLA-4.
Data processing |
The microarray data were preprocessed in R employing the RMA algrorithm. A custom CDF file was used (brain array; mogene10stmmentrezgcdf_18.0.0).
