Treatment protocol For treated group, Cells were incubated to an OD600 of approximately 0.8, centrifuged and resuspended in phosphate buffer, then treated with 100 mM H2O2 for 30 minutes.
Growth protocol D. radiodurans R1 and its derivatives were grown at 30°C in TGY broth (0.5% tryptone, 0.1% glucose, and 0.3% yeast extract).

Extraction protocol RNA was harvested using Trizol reagent. 4 µg of total RNA was used for the construction of sequencing libraries.

Sequenced reads were trimmed for adaptor sequence, and masked for low-complexity or low-quality sequence, then mapped to R1 whole genome using soap maximum number of mismatches allowed = 2bp, number of valid alignments per read pair (-k) = 40
Fragments Per Kilobase of exon model per Million mapped reads (FPKM) were calculated.
