Treatment protocol siRNA pools for Pumilio 1 and Pumilio 2 (siRNAs from Dharmacon/GE) were mixed and diluted to a final concentration of 50nM and transfected overnight in a 24-wells culture plate.
Growth protocol HEK293T cells were maintained at 37⁰C at 5% CO2 in Dulbecco's Modified Eagle's Medium (DMEM), high glucose (4500mg/L), supplemented with Penicillin (100units/ml), Streptomycin (100μg/ml), Glutamine (2mM) and 10% heat-inactivated fetal calf serum.
Extracted molecule total RNA
Extraction protocol RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.

Hybridization protocol Standard Illumina hybridization protocol.
Scan protocol Arrays were scanned using the iScan system (Illumina, Inc.), and the fluorescence intensities were analyzed and annotated using GenomeStudio software (Illumina, Inc.)
