siRNA pools for Pumilio 1 and Pumilio 2 (siRNAs from Dharmacon/GE) were mixed and diluted to a final concentration of 50nM and transfected overnight in a 24-wells culture plate.

Growth protocol |
HEK293T cells were maintained at 37⁰C at 5% CO2 in Dulbecco's Modified Eagle's Medium (DMEM), high glucose (4500mg/L), supplemented with Penicillin (100units/ml), Streptomycin (100μg/ml), Glutamine (2mM) and 10% heat-inactivated fetal calf serum.

Extraction protocol |
RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.
