An immunoglobulin, utilized as inner carrier, was labeled with a C5y monoreactive dye and incubated with the viral protein and protein A microbead to form multivalent complexes. These complexes were supplemented with soluble protein A microarray to block non-specific binding to the protein microarray slides.

Hybridization protocol |
Array slides were loaded onto an automated a-Hyb hybridization station, blocked with 5% milk in PBS.

Scanned with a GenePix 4000B scanner. GenePix Pro 6.0 software was used for analysis.

Data processing |
The protein microarray analysis was done with a z-score based on the array distribution and scores were standardized with quantile normalization. Probes that bound to 5% of more of the baits were flagged as non-specific binders, as determined by a proportion test of a representative set of screens. After eliminating non-specific binders, probes with a score of 5.0 or higher were counted as hits. Please see Methods section in the accompanying paper for further details on experimental protocol and data analysis. Given the number of human proteins in the collection, samples were randomly distributed in 2 libraries, called 'Array 1' and 'Array 2'. To control for slide variability, duplicate microarrays from separate spotting runs were analyzed in all cases. The average score from the duplicate runs for each of the viral proteins screened is provided in the Matrix table. Probes with a score of 5 or higher were counted as hits.
