A single pair of gonads (14.5 dpc) was separated from the mesonephros and was used as one biological replicate (4 biological replicates were included for each genotype). Total RNA was isolated using Picopure extraction kit (Arcturus). RNA quality and concentration were determined using N Agilent 2100 BioAnalyzer (Agilent Technologies, Santa Clara, California)

One hundred nanograms (100 ng) of total RNA were amplified and labeled as directed using the Affymetrix WT Plus Reagent Kit (WT Plus Kit).

Hybridization protocol |
5.5 μg micrograms of amplified biotin-cDNAs were fragmented and hybridized to each array for 16 hours at 45°C in a rotating hybridization oven. Array slides were stained with streptavidin/phycoerythrin then washed for antibody amplification according to the GeneChip Hybridization, Wash and Stain Kit and user manual protocol FS450-0001.
