Biotin labeled cRNA targets were prepared from 6.2ug total RNA using Affymetrix's single-cycle eukaryotic target preparation procedure (Expression Analysis Technical Manual, 2001, Affymetrix). Reverse Transcription was performed using Superscipt II Choice System (Invitogen). Invitro-Transcription and labeling was conducted using the GeneChip Expression 3' Amplification Reagent Kit (Affymetrix).

Hybridization protocol |
Following fragmentation, 15ug of biotin labeled cRNA was hybridized for 16hr 28min at 45C, 60rpm in an Affymetrix Hybridization Oven 640 to a Rat230 2.0 GeneChip expression array. The washing and antibody amplification staining procedure utilizing R phycoerthrin-streptavidin was performed with the Affymetrix Fluidics Station 450 using the EukGE-WS2v5_450 fluidics protocol.
