UET-13 transfectants were treated with or without 3 μg/ml of tetracycline for the indicated periods.

Growth protocol |
UET-13 transfectants (UET-13TR-EWS/FLI1 and UET-13TR-EWS/ERG ) were cultured in Dulbecco’s modified Eagle’s medium (DMEM) with 10% Tet System Approved Fetal Bovine Serum (T-FBS)(Takara, Shiga, Japan) at 37°C under a humidified 5% CO2 atmosphere.

Total RNA was extracted from cells using RNeasy kit (QIAGEN, Valencia, CA) according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA.

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix HG-U133 Plus 2.0. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneArray Scanner GCS3000.

Data processing |
The arrays were analyzed using GeneChip Operating Software 1.2 (GCOS 1.2) (Affymetrix). Background subtraction and normalization were performed by GeneSpring GX 7.3 software (Agilent Technologies, Inc., Santa Clara, CA).
