Growth protocol In vitro-fertilized embryos derived from wild-type and miRNA-deleted mice were cultured in KSOM medium at 37°C for 16 hours. Cultured embryos were transferred into pseudopregnant ICR females and E11.5 embryos were retrieved from the uterus.
Extracted molecule total RNA
Extraction protocol Placentas with deciduas removed were lysed and the total and miRNA isolated using a mirVana miRNA isolation kit (Themo Fisher Scientific).
Label Cy3
Label protocol For miRNA analysis, total RNA from placentas was labeled with the miRNA Complete Labeling Reagent and Hyb Kit (Agilent Technologies). For mRNA analysis, total RNAs were labeled with a Low Input Quick Amp Labeling Kit (Agilent Technologies).
 
Hybridization protocol For miRNA analysis, Cy3-labeled RNAs was hybridized with the miRNA Complete Labeling Reagent and Hyb Kit onto mouse miRNA Microarray rel.17.0 (Agilent Technologies) at 65°C for 17 hours according to manufacture's instructions. For mRNA analysis, 0.6ug Cy3-labelled RNAs were fragmented at 60°C for 30 minutes and hybridized onto SurePrint G3 Mouse GE microarray 8x60K (Agilent Technologies) at 65°C for 17 hours according to manufacture's instructions.
Scan protocol Scanning of microarray slides was performed with a DNA microarray scanner at 3 um resolution (Agilent Technologies).

Data processing Scanned image files were processed to give signal intensities using Feature Extraction software (Agilent Technologies). miRNA raw intensity signals were not normalized.
