Sample type |
RNA |
Source name |
Primary monocytes cultured in normal glucose, IFNgamma
Organism |
Homo sapiens |
Characteristics |
patientno: 4 treatment: IFNg glucose: NG cell type: monocyte-derived macrophages cell subtype: M1
Treatment protocol |
After isolating monocytes from buffy coats, these cells were cultured with IFNg (100ng/ml), IL4 (10ng/ml) and control (without cytokine) for 6 days
Growth protocol |
Primary human monocytes were cultured in serum free medium supplemented by M-CSF (5ng/ml) for 6 days with specific cytokine stimulations in order to differentiate in to different sub-types of macrophage population (M0,M1 and M2).
Extracted molecule |
total RNA |
Extraction protocol |
RNA was isolated by using E.Z.N.A. total RNA kit. Extraction procedure was according to E.Z.N.A. total RNA kit
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol.
Hybridization protocol |
Hybridization (16h x 45°C) was processed according to the standard Affymetrix protocol.

Scan protocol |
Affymetrix GeneArray Scanner3000
Description |
Monocytes cultured in normal glucose (5mM) for 6days, IFNgamma (100ng/ml) Normal_IFNg_4
Data processing |
The data were analyzed with a commercial software called JMP Genomics, version 7, from SAS. Gene expression profiling was performed using arrays of human Hugene-1_0-type from Affymetrix. A Custom CDF Version 19 with Entrez based gene definitions was used to annotate the arrays. The Raw fluorescence intensity values were normalized applying quantile normalization, RMA background correction and Medianpolish Probeset Summary
