Title |
Shoot control rep1 |
Sample type |
RNA |
Source name |
Arabidopsis shoots
Organism |
Arabidopsis thaliana |
Characteristics |
Arabidopsis thaliana, ecotype WS
Biomaterial provider |
RIKEN Institute, Yokohama, Japan
Treatment protocol |
Root samples were transferred to liquid nitrogen before RNA extraction.
Growth protocol |
WS plants were grown at 22C under 16h light and 8 hour dark cycle.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was prepared using RNeasy Plant Mini Kit (Qiagen, Hilden, Germany) according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 10 microg total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
Hybridization protocol |
Following fragmentation, 15 microg of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Gene Array Scanner (Agilent Technologies, Palo Alto, CA).
Description |
Gene expression data from roots of plants vertically grown for 10 days on 0.5 MS media.
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
