Treatment protocol |
Adult flies of the above genotypes were subjected to a 1hr heatshock at 37C and then allowed to recover for 3hrs at 25C.

Adult male and female flies were raised at 25C. W1118 was used as a control. HspGal flies were crossed to flies carrying one of the folowing transgenes, UAS-EYA-wildtype, UAS-EYA-D493N or UAS-EYA-E728Q and flies carrying both the driver and UAS-EYA trangene were collected

Extraction protocol |
Flies were transferred to falcon tubes, frozen in liquid nitrogen and vortexed, This was repeated two more times and fly heads were collected using a paint brush from an Rnase-free plate. RNA was isolated from 200 adult heads (4 groups of 50) in Trizol according to the manufacturer's protocol and RNA was cleaned up using the Qiagen RNeasy kit.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 2 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
Chips were scanned using the GeneChip Scanner 3000 enabled for High-Resolution scanning.

GeneChip Operating Software (GCOS) v1.1.1. Global mean normalization was performed for samples, scaled so that each median is 100. Detection P values were determined from raw data using GCOS software.
