N/A
Growth protocol |
Bone-marrow-derived human mesenchymal stem cells (Bm MSC, Texas A&M University System Health Science Center), adipose-derived human MSC (Ad MSC, Thermo Fisher Scientific) and human neonatal dermal fibroblasts (NHDF, Lonza) were seeded on each substrate (i.e., Bm ECM, Ad ECM, Der ECM or TCPS) at a cell density of 3,500 cells/cm2. Cells were cultured in α–MEM medium supplemented with 20% FBS and 1% penicillin-streptomycin for 3 days.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted from cell pellet using RNeasy Mini Kit (Qiagen, Hilden, Germany) following manufacturer's instructions
Label |
biotin
Label protocol |
Total RNA was amplified and labeled GeneChip WT Plus Reagent Kit (Affymetrix)
Hybridization protocol |
5.2ug of ss-cDNA is hybridized 16h at 45C GeneChip® PrimeView™ Human Gene Expression Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the 7G Gene Chip Scanner
Description |
Gene expression data from Bm MSC on Ad MSC-derived matrix
Data processing |
CEL files were processed, summarized and normalized using R/Bioconductor packages affyPLM_1.46.0, preprocessCore_1.32.0 and affy_1.48.0 using RMA.
