Title |
Naïve T-injected severely inflamed colon tissue, biological rep2 |
Sample type |
RNA |
Source name |
colon tissue from Naïve T-injected mice
Organism |
Mus musculus |
Characteristics |
strain: C57BL/6 genotype: Rag1-/- treatment: Naïve T-injected tissue: severely inflamed colons age: 4-5 mos
Treatment protocol |
Colons were excised and distal colon tissue (from rectum to proximal/distal junction) was snap frozen.
Growth protocol |
Animals were injected with Tregs or naïve T cells at 10 weeks of age and sacrificed 2-4 months after injection.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted using the Qiagen RNAeasy tissue microarray tissue mini kit following manufacturers recommended protocol.
Label |
biotin
Label protocol |
The targets were prepared using the Affyemtrix GeneChip WT PlUS Reagent Kit and labeled by biotinylation.
Hybridization protocol |
Following labeling, biotinylated cRNA was hybridized to the GeneChip® Mouse Exon 1.0 ST Array for 16 hr at 45C in an Affymetrix HybridizationOven. Hybridized array was washed and stained using fluorescent stain SAPE and Anti-streptavidin antibody.
Scan protocol |
The arrays were scanned on GeneChip Scanner 3000 7G
Description |
Gene expression data from severely inflamed colons
Data processing |
The data were extracted and quantified at the gene level using the Affymetrix expression console with RMA processing.
