Growth protocol |
Sterilized seeds of transgenic plants and wild type were germinated in MS medium. Positive transgenic plants were selected by adding 50 mM hygromycin for S58S, or 50 mM G418 for S59R into the medium. Ten-day-old uniform seedlings were transferred to hydroponics culture medium and 1-month-old whole plants were collected for microarray analysis. All transgenic plants were prechecked by real time RT-PCR for the expression level of OsSKIPa to confirm overexpression or suppression of the gene.
