Title |
large airways, smoker 161 |
Sample type |
RNA |
Source name |
large airway epithelial cells obtained by bronchoscopy and brushing
Organism |
Homo sapiens |
Characteristics |
Age: 35 Sex: M Ethnic group: black Smoking status: smoker, 5 pack-years
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction and RNAeasy clean-up of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 microg total RNA for HuGeneFL and HG-U133A or 2 microg total RNA for HG-U133 Plus 2.0 (Expression Analysis Technical Manual, 701022 Rev.2, Affymetrix).
Hybridization protocol |
Following fragmentation, 15 microg of cRNA were hybridized for 16 hr at 45C on GeneChip HuGeneFL, HG-U133A or HG-U133 Plus 2.0. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using GeneArray 2500 for HuGeneFL and the GeneChip Scanner 3000 7G for HG-U133A and HG-U133 Plus 2.0.
Description |
Comparison of gene expression in large airway epithleium of normal non-smokers and normal smokers
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method.
