Cord blood CD34+ cells were grown in Stemline II medium supplemented with TPO, SCF and Flt-3L, cells were subjected to 2 rounds of lentiviral transduction at MOI 4-8. Cells were harvested for gene expression profiling 24 hours after the last transduction.

Extraction protocol |
RNA was prepared using the Trizol Reagent (Invitrogen, Carlsbad, CA) and RNeasy micro kit (Qiagen, Valencia, CA) following the manufacturer's recommendations. The RNA quality and integrity were analyzed with Nanodrop ND 1000 spectrophotometer (Rockland, DE) and Bioanalyzer (Agilent, Cheshire, UK).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100ng total RNA (Two-Cycle cDNA Synthesis Kit and IVT labeling kit).

Hybridization protocol |
Following fragmentation, 20 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix Human Genome Gene v2 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400 (EukGE-Ws2v4).

Scan protocol |
GeneChips were scanned using the GeneChip Scanner 3000 7G (Affymetrix).
