Source name |
ExVivoColon_DNBS+L. rhamnosus
|
Organism |
Mus musculus |
Characteristics |
tissue: Colon age: 10w gender: Male strain: C57BL/5 treatment: DNBS+L. rhamnosus replicate: 3
|
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was prepared from total tissue of colon by using the column-based RNeasy® Micro Kit (Qiagen). RNA was quantified using a NanoDrop-1000 spectrophotometer and RNA quality was monitored with the Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA). Samples with RNA integrity number (RIN) ranging from 8.5-10 were selected for further analysis.
|
Label |
Cy3
|
Label protocol |
Labelling of total RNA (100ng) was performed with the One-Color Low Input Quick Amp Labelling kit (Agilent Technologies).
|
|
|
Hybridization protocol |
Samples were hybridized to SurePrint G3 mouse 8x60K v2 (G4858A-074809) arrays and scanned by G2565CA Scanner System (Agilent Technologies).
|
Scan protocol |
Slides were scanned immediately after washing on the Agilent DNA Microarray Scanner (G2505C) using one color scan setting for 1x60k array slides (Scan Area 61x21.6 mm, Scan resolution 10um, Dye channel is set to Green and Green PMT is set to 100%).

Description |
Gene expression after 23 days of treatment by DNBS+L. rhamnosus Sample name: 5.4
|
Data processing |
Image analyses and raw signal intensities were quantified by Feature Extraction Software v10.7.3.1 (Agilent Technologies) according to the GE1_107_Sep09 protocol.
