Treatment protocol |
A549 cells were used to infect with influenza H1N1 virus (BJ501). A549 cells were washed and incubated with virus at a multiplicity of infection (MOI) of 1 for 1 h with FBS-free medium . After adsorption, the supernatant was aspirated and cells were cultured in medium supplemented with 2% FBS and 100 units penicillin-streptomycin ml-1 at 37°C in a humidified atmosphere of 5% (v/v) CO2 for indicate hours. 12 h with IAV WSN a nd SeV, o r 24 h with MDRV and HSV.

Growth protocol |
A549 cells were purchased from the ATCC and cultured in DMEM/F-12(1:1) basic(Gibco) supplemented with 10% FBS and 100 units penicillin-streptomycin ml-1 at 37°C in a humidified atmosphere of 5% (v/v) CO2.

About 1ug total RNA were polyA tailed and labled with FlashTag™ Biotin HSR Labeling Kit. This kit can be used to label any RNA sample, including total RNA, severely degraded RNA, plant RNA, and low molecular weight RNA.

Hybridization protocol |
The labled RNA were hybridized for 16 hr at 48C on Affymetrix miRNA Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip® Scanner 3000.

The data were analyzed with miRNA QCTool using Affymetrix default analysis settings and quantile as normalization method.
