Treatment protocol |
Third instar larvae were collected for each genotype and a total of 50 larvae for each replicate were used to proceed with Trizol RNA extraction.

Growth protocol |
To induce innate immune responses, we generated a transgenic Drosophila line that allows cell-restricted activation of Imd and crossed it with a fat body specific driver (R4-GAL4). Larvae from R4-GAL4/imdca group and control genotypes (CA/+ and R4-GAL4/+) were raised on standard food at 25°C until they reached third instar stage and then collected for RNA extraction.

Biotinylated cRNA were prepared according to the standard Affymetrix GeneChip 3' IVT plus protocol from 100 ng total RNA (User Guide GeneChip® 3' IVT PLUS Reagent Kit, 2013-2015, Affymetrix).

12 ug of cRNA was used for fragmentation, fragmented cRNA was hybridized on GeneChip®Drosophila Genome 2.0 Array, washed and stained according to the Affymetrix protocol (User Guide GeneChip® 3' IVT PLUS Reagent Kit, 2013-2015, Affymetrix).
