Treatment protocol |
hPS were plated at approximatively at 30,000 cells/cm2 in mTeSR, and after recovery medium was changed to a DMEM-based medium supplemented with insulin-transferrin- selenium (ITS, Gibco), 3 μMCHIRON99021 (Axon MedChem, Tocris) (C medium) and additionally 0.5 μM LDN-193189 (Axon MedChem, Stemgent) (CL medium). On day 3, 20 ng/ml FGF2 (R&D Systems) was added .

Extraction protocol |
Total RNA was extracted from PSC populations using Trizol (Invitrogen) or with the RNAeasy plus mini-kit (Qiagen).

Biotinylated cRNA targets were prepared from total RNA using a double amplification protocol according to the GeneChip® Expression Analysis Technical Manual: Two-Cycle Target Labeling Assay (P/N 701021 Rev.5, Affymetrix, Santa Clara, USA).

Hybridization protocol |
Following fragmentation, cRNAs were hybridized for 16 hours at 45oC on GeneChip® Human Genome arrays. Each microarray was then washed and stained on a GeneChip® fluidics station 450.

Scan protocol |
GeneChips were scanned using the GeneChip® Scanner 3000 7G. Raw data (.CEL Intensity files) were extracted from the scanned images using the Affymetrix GeneChip® Command Console (AGCC) version 3.1.
