Treatment protocol |
Splenocytes were isolated and antigen-specific CD4+ T cells were enriched using Biotin-labeled anti-CD19, anti-Ter119, anti-CD8, anti-CD11c, anti-CD11b, anti-NK1.1, anti-CD25 and magnetic bead purification (Beaver). Virus specific SMtg CD4+CD45.1+CD19-L/D- cells were then sorted into TFH and TH1 populations on the basis of CXCR5 and SLAM expression using a FACS Aria II (BD Bioscience).

Growth protocol |
all infection were done by intraperitoneal injection of 2 x 10^5 plaque-forming units (PFU) of LCMV Armstrong strain per mouse. 2 x 10^5 naïve SMARTA CD4+T cells were adoptively transferred to recipient mice prior to LCMV infection. Spleens were harvested 6 days after LCMV infection.

Sorted cells were lysed with TRIzol LS reagent (Life Technologies). Total RNA was extracted with isopropyl ethanol.

RNA quality of all samples was confirmed by BioAnalyzer Nano gel (Agilent), then probes were generated from 30ng total RNA by single round linear amplification using the Ovation Pico system (Nugen).
