Growth protocol |
Sterilized seeds of transgenic plants and wild type were germinated in MS medium. Positive transgenic plants were selected by adding 50 mM hygromycin for miR156-overexpressed plants. Ten-day-old uniform seedlings were transferred to hydroponics culture medium and forty five-day-old plants were collected for microarray analysis. The leaves in the main tiller were numbered according to its emerging order, with L1 indicating the first developed leaf. All samples were prechecked by small RNA gel blot for the expression level of miR156 to confirm overexpression. The old (L3) and young leaf (L6) were used for microarray hybridization.
