Extraction protocol Progenitor cells at each stage were sorted by a combination of cell surface markers for IL-3R, CD34 and CD36. Erythroblasts at each stage of terminal erythroid differentiation were sorted by a combination of cell surface markers for glycophorin A, band 3, and α4 integrin. Genomic DNA was isolated using a Qiagen Gentra Puregene kit.
Enhanced Reduced Representation Bisulfite sequencing (ERRBS) was performed at the Weill Cornell Genomics Core.

Data processing Fastq format reads were trimmed using Trim Galore v0.4.0 in rrbs mode

Differential methylation was identified using methylKit v1.2.4
