Bone marrow from female BXD recombinant inbred mice between 3 and 6 months of age were flushed from the femurs and tibiae of three mice and pooled, for each sample. After standard erythrocyte lysis nucleated cells were were stained with a panel of biotinylated lineage-specific antibodies (Mouse Lineage Panel, containing anti-CD3e, anti-CD45R/B220, anti-CD11b (Mac-1), anti-TER119 (Ly-76), anti-Gr-1 (Ly-6G), Pharmingen, San Diego, CA), FITC-anti-Sca-1 and APC-anti-c-kit (Pharmingen). Stem cell yield across all BXD samples varied from 16,000 to 118,000 Lin-Sca-1+ c-kit+ cells. Cells were washed twice, and incubated for 30 minutes with streptavidin-PE (Pharmingen). After two washes cells were resuspended in PBS with 0,2% BSA, and purified using a MoFlo flowcytometer. The lineage-depleted bone marrow cell population was defined as the 5% cells showing least PE-fluorescence intensity. Cells was immediately collected in RNA lysis buffer.
