Treatment protocol IVF or SCNT embryos were retrieved from the mother’s uterus at E11.5 and placentas without deciduas were used for RNA extraction.
Growth protocol 1-cell stage embryos derived from IVF and SCNT were cultured in KSOM medium at 37°C for 16 hours. Cultured embryos were transferred into pseudopregnant ICR females and 11.5 embryos were retrieved from the uterus.
Extracted molecule total RNA
Extraction protocol Total RNA from E11.5 placentas was extracted from IVF and SCNT placentas using RNeasy Mini kits (QIAGEN).
Label Cy3
Label protocol Total RNA was labeled with miRNA Complete Labeling and Hyb Kit (Agilent), according to the manufacturer’s instructions.
 
Hybridization protocol Labeled RNA was hybridized to SurePrint G3 Mouse miRNA 8x60K (Agilent) at 55˚C for 20 hrs, according to manufacture's instructions.
Scan protocol Scanning of microarray slides was performed on a DNA microarray scanner (Agilent) at a 5 um resolution.
Description miRNA expression of E11.5 SCNT placentas
Data processing Scanned-image files were processed to give signal intensities using the Feature Extraction software (Agilent). All raw data were loaded into Gene Spring GX software (Agilent) and 90 percentile shift was performed.
Raw intensities after 90 percentile shift. Signals under limit of detection were set as 0.1.
