Growth protocol |
OG2MEF cells were transduced twice by pMX-Sox2, Klf4 along with OCT4, SOX17 or SOX17-V118M or GFP retroviruses produced using PlatE cells in DMEM + 10% FBS media supplemented with 1x NEAA and Glutamax. After second viral transduction, the medium was switched to mouse ESC media ((DMEM containing 4.5 g/L D-glucose, 15% FBS, 1% NEAA, 1% GlutaMAX, 1% Sodium Pyruvate, 0.5% penicillin/streptomycin, 1000 U/mL Leukemia inhibiting factor (LIF, prepared by the core facility of GIBH), 0.055 mM β-ME (beta mercaptoethanol) and 50 μg/mL Vitamin C).
