Title |
Peripheral blood monocytes, CD16 negative, rep4 |
Sample type |
RNA |
Source name |
peripheral blood monocytes, CD16-
Organism |
Homo sapiens |
Characteristics |
donor health: healthy tissue: peripheral blood cell type: monocyte cd16: negative
Treatment protocol |
ex vivo
Growth protocol |
ex vivo
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA from Mo pellets was isolated by Trizol extraction and purified using RNeasy columns (Qiagen). The quality of RNA was assessed by visualization of intact bands corresponding to 18S and 28S rRNA on formaldehyde agarose gels.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 10 ug total RNA (Expression Analysis Technical Manual, 2003, Affymetrix).
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Human Genome U133 Plus 2.0 Array (Platform ID: GPL570). GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner 2500A.
Description |
Gene expression data ex vivo. 042304_CD16_negative
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
