Status Public on Jun 16, 2020
Title BT2 hTSC Rep2
Sample type SRA

Source name BT2 hTSC
Organism Homo sapiens
Characteristics cell type: Human trophoblast stem cells
Growth protocol hTSCs were cultured according to published protocol(Okae et al., 2018) with the following alterations. 0.5-1 x 105 cells were plated on each well of a six-well plate, and cells were passaged every 5-7 days. Cells were grown at 5% O2 5% CO2
Extracted molecule total RNA
Extraction protocol RNA extraction was performed using Qiagen RNeasy micro kit
mRNA was enriched from 500ng total RNA using NEBNext Poly(A) mRNA Magnetic Isolation Module kit and libraries were generated using Swift RNA library kit

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NovaSeq 6000

Data processing FASTQ files were mapped to hg19 using the STAR aligner with default parameters.
Bam files were analyzed by RNA-SeQC to confirm library quality.
Read counts were calculated with htseq-count and RPKM was calculated using cufflinks with default settings.
