Growth protocol Cells were grown in high glucose DMEM with Glutamax, supplemented with 10% FBS and antibotics; six different passages were collected in a paired fashion (one sample for each cell type) ~ 24 h after plating, pelleted and frozen at -80 C until tRNA extraction.

Extraction protocol tRNA was extracted using Quiagen RNeasy kit with on column DNAse digerstion according to the manufacturer's instructions.

Label protocol Biotinylated cRNA was prepared using Ambion MessageAmp II-Biotin Enhanced Kit according to the manufacturers's instructions, starting from 1 ug of tRNA/sample.
