Growth protocol |
Wt and p27delta51 MEFs were cultured in DMEM supplemented with 10% fetal bovin serum (FBS). All cultures were maintained at 37ºC and 5% CO2.

Extraction protocol |
Total RNA was extracted using the RNeasy Kit (QIagen). RNAs from quiescent MEFs from wt or delta51 mice were analyzed using a Bioanalyzer (Agilent). Only samples with RIN number greater than 9 and a ratio A260/280 over than 2 were further used in microarray analysis. Retrotranscription was made followed by labeling with biotin.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol

Hybridization protocol |
Following fragmentation, cRNA were hybridized for 16 hr at 45ºC on Affymetrix 430 2.0 murine array. Chips were washed and stained in the Affymetrix Fluidics Station.

Arrays were scanned using the Scanner Axon 4000B

Data processing |
The normalization of the arrays was done using the rma method as implemented in the Affy package of R Bioconductor. Quality control measurements were done using ArrayQualityMetrics package of R Bioconductor.
