Title |
UM-PEL-1 xenografts, treated with Bortezomib; material pooled from three mice |
Sample type |
RNA |
Source name |
UM-PEL-1 tumor xenografts in NOD/SCID mice
Organism |
Mus musculus |
Characteristics |
lymphoma type: Primary Effusion Lymphoma cell line: UM-PEL-1 agent: Bortezomib
Treatment protocol |
Mice were injected IP a single time with either PBS or bortezomib (0.3 mg/kg). Cells were recovered from each of the three mice per treatment group after 24 hours and pooled.
Growth protocol |
UM-PEL-1 cells, that were not cultured in vitro, were injected into the peritoneal cavities of NOD/SCID mice and grown for 3 days prior to treatment.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was isolated by TRIzol reagent according to the manufacturer's insturctions.
Label |
biotin
Label protocol |
RNA was amplified and labeled using NuGEN Ovation Pico WTA system, WT-Ovation Exon module and Encore Biotin Module (NuGEN, San Carlos, CA, USA).
Hybridization protocol |
cDNA was used to generate sense transcript cDNA used for hybridization on the Affymetrix Human Gene 1.0ST arrays.
Scan protocol |
According to Affymetrix protocol.
Description |
UM-PEL-1 + Bortezomib
Data processing |
Image analysis was done using the Affymetrix Command Console Software (AGCC). Resulting CEL files were be imported into Partek software (Partek, Inc., St. Louis, MO) using median polish probeset summarization, GCRMA background correction and quantile normalization.
